2.6 Calibration Curve of Standard

Description: A dilution series was done on the internal standards with concentrations ranging from 50 fmol to 0.1 fmol. Peptide sensitivity was reliable down to 0.1 fmol.

 

Experiment Description: Cells from 8226 line were lysed in RIPA buffer. The equivalent of 100,000 cells were loaded onto an SDS gel. The protein of interest was excised and ingel digestion with trypsin was performed after reduction and alkylation with TCEP and IAA. 1/4 of the resulting digest was then analyzed on a Thermo Scientific TSQ mass spectrometer.
